Here, all process steps ofthe established method for purification ofthe enzyme urease from jack beans are analyzed and optimized in terms ofthe product yield, urease purity, and crystal size. The purification process consists oftwo main steps: extraction ofurease from the jack bean meal and cooling crystallization, each followed by solid-liquid separation. As separation procedures both centrifugation and filtration were tested for the extraction as weil as for the crystal separation. Furthermore, a set ofexperiments was carried out, where an intermediate precipitation ofproteins from the extract before urease crystallization was performed. As final purification steps recrystallization and crystal washing were carried out. Additionally, the optimum urease storage parameters were investigated.
