Sugarcane is the major crop of Pakistan. Near about 90 industries are run on sugarcane. The main issue of sugarcane in Pakistan is water deficit and viable FUZZ production. The water issues are going increase day by day and the production level of sugarcane is decreased. The ecosystem of Pakistan is not suitable for the production of viable FUZZ therefor, the true breeding of sugarcane in Pakistan is a challenge. Different options are going to practice for the new lines and varieties, including tissue culture techniques, somaclonal variations, introduce new germplasm. For the solution of like these problems, we made some experiments in this regard by applying polyethene glycol (PEG) as in-vivo and in-vitro. After applying PEG, made experiments on their leaves to check their strength against drought in in-vivo experiments. For checking drought strength I used proline, glycine betaine, phosphomolybdenum, DPPH, phenolics, flavonoids and DNA denature. The same study was done by using in-vitro techniques. For DNA studies different primers were used to check the similarity index between the species of sugarcane. Our findings were showed optimized results for the production of sugarcane.
